The epicenter of the tremor, which occurred at 4.28am (2328 GMT Saturday), was located at 99 kilometres (61 miles) north of Kulob at a depth of 10 kilometres, USGS said.
There were no immediate reports of casualties or damage in the Kulob district which is 200 kilometres southeast of the capital Dushanbe.
The reading was based on the open-ended Moment Magnitude scale, now used by US seismologists, which measures the area of the fault that ruptured and the total energy released.
