The 44-year-old, who has not been named, is accused of raping minors four times in the US state of Kentucky in 2003, a Seoul Metropolitan Police Agency (SMPA) spokesman said.
"He will soon be extradited to the US," he said.
The suspect, who arrived in South Korea in 2004 via Thailand, has been working as an English teacher in the southwestern province of North Jeolla.
South Korean police launched a search for the suspect in February at the request of US authorities.
