Roddick, a former world number one and the 2003 US Open winner, had never got beyond the fourth round at the French Open in nine visits.
He had been defeated in his opening matches at the claycourt Madrid and Rome Masters this month.
Spain's Juan Carlos Ferrero, the 2003 French Open champion, also announced he had pulled out of the second Grand Slam event of the season which starts on Sunday.
Ferrero, 31, has played just two tournaments this year, both in his native Spain, as his ranking has slipped to 72.
