Johansson, playing at Flushing Meadows for only the second time, took his ace tally for the tournament to 76 as he overwhelmed Llodra on Grandstand Court.
The 28th seed dropped only his second service game of the tournament in the second set but he recovered quickly to set up a last-eight meeting with second seed Andy Roddick, who put out Spain's Tommy Robredo 6-3 6-2 6-4.
Johansson is the first Swede to reach the quarter-finals at the US Open for four years.
