The accident occurred on Saturday at Haripur in Sunsari district on the East-West Highway, police said.
They said the bus, which was heading towards Janakpur from Bhadrapur in eastern Nepal, veered off the road and met with the accident, killing five people and injuring 55.
The injured are being treated at BP Koirala Institute of Health Sciences in Dharan and Koshi Zonal Hospital, police said.
Separately, a motorcyclist was killed when a tanker hit him in Birgunj in southern Nepal, while another person died in a truck-bus collision in Nawalpwarsi, they said.
A police constable was killed in another road accident involving a bus and a truck in Dhanusha district in southern Nepal.
