Shubhranjan Sen, field director of Pench tiger reserve, said Gunatrao, Kamal and Sunil -- all residents of Turiya village in Seoni district -- were arrested for poisoning the tigress and her cubs.
The tigress, popularly called the Baghin nala female, was found dead on March 28 and two of her four cubs were found dead on March 29.
The park management also found the body of a spotted deer near the same spot which had been partially eaten by the tigress.
The park management suspects that the villagers killed the spotted deer by poisoning it and the tigress died as she ate the deer.
However, no concrete evidence linking the accused to the tigress killing has been found by the forest department.
Sources in the forest department said they had found evidence that linked the accused to the crime but they were not willing to make it public.
