The bomber blew up an explosives-filled minivan at the checkpost in Nangarhar province, governor Gul Agha Sherzai told reporters.
"There was a suicide attack in Chaparhar district in which one policeman and three civilians died," he said.
The attack took place about 30 kilometres (20 miles) south of the provincial capital Jalalabad, an AFP reporter said. The bomber had apparently been trying to enter the bustling city.
There was no immediate claim of responsibility for the attack but the mounting Taliban-led insurgency in Afghanistan has been marked by a wave of such bombings.
