The 42-year-old will be seen playing a sex-obsessed, kinky dentist in Horrible Bosses, reported Sun online.
Her role in the film is said to be a "total departure" from her usual romantic comedy characters.
Aniston has previously only appeared partially nude in a blurred scene in the 2007 film The Break-Up.
"It was worked out before she signed her contract, so she's committed," said a source on the sets of the film. The actress has never stripped on screen, but has posed for magazines with her clothes off.
Last year, to celebrate her 40th birthday, Aniston had posed naked for GQ magazine with only a tie to cover her modesty.
